The Influence of Al Ions on Pepsin and Trypsin Activity in Vitro
نویسندگان
چکیده
The objective of this study was to investigate the influence of the concentration of Al ions and the substrate/enzyme ratio on pepsin and trypsin activity in vitro. The experimental design was a combination of three Al ion concentrations (0.25; 2.5 and 25.0 μg Al/ml of reaction solution) and two substrate/enzyme ratios (S/E = 10 and 100 for pepsin, and 100 and 1000 for trypsin). Enzymatic activity was determined by the Folin method based on the reaction of tyrosine with the Folin reagent. It was found that the concentration of Al ions influenced activity of pepsin which increased with the increasing metal ion concentration in the reaction milieu. Al ions did not affect activity of trypsin. Proteolitic activity of pepsin and trypsin depended on substrate/enzyme ratio. Higher concentration of substrate decreased efficacy of enzymatic protein breakdown in vitro.

Bioaffinity Based Immobilization of Almond (Amygdalus communis) b-galactosidase on Con A-layered Calcium Alginate-cellulose Beads: Its Application in Lactose Hydrolysis in Batch and Continuous Mode
In this study, immobilization of partially purified almond (Amygdalus communis) β-galactosidase on Con A layered calcium alginate-cellulose beads was investigated. Immobilized β-galactosidase retained 72% of theinitial activity after crosslinking by glutaraldehyde. Both soluble and immobilized enzyme exhibited the samepH and temperature optima at pH 5.5 and 50ºC, respectively. Howev...
متن کاملThe Coating Effect of PANI/Silver on Performance of Polysulfone Membrane Toward Protein Separation
The effects of different coating time of PANI/Silver onto polysulfone (PSf) membrane surface were investigated based on the morphology, contact angle, surface roughness and rejection towards BSA, pepsin and trypsin. The membrane was prepared by employing the pressure deposition method toward phase inversed membrane. Thus, PANI particles were forced to adhere on membrane surface by pressure driv...
متن کاملScreening of whey protein isolate hydrolysates for their dual functionality: influence of heat pre-treatment and enzyme specificity.
Heat pre-treated and non heat pre-treated whey protein isolate (WPI) were hydrolysed using α-chymotrypsin (chymotrypsin), pepsin and trypsin. The in vitro antioxidant activity, ACE-inhibition activity and surface hydrophobicities of the hydrolysates were measured in order to determine if peptides with dual functionalities were present. Dual functional peptides have both biological (e.g. antioxi...

The purpose of this study was to survey the effect of dietary fish oil replacement with different levels of soybean lecithin on growth performance and digestive enzymes activity of Persian sturgeon. Three hundred and sixty fish with average weight of 85 ± 0.09 g were treated with 6 experimental diets and three replicates, containing 0, 2, 4, 6, 8 and 10 percent soybean lecithin for 10 weeks. Ba...
متن کاملThe Mechanism of the Influence of Acids and Alkalies on the Digestion of Proteins by Pepsin or Trypsin. By
I t has long been known that pepsin digestion occurs only in acid solution while trypsin digestion takes place in neutral or alkaline solutions. This marked influence of the reaction on the rate of hydrolysis is a characteristic property of enzymes in general. A great advance in the explanation of this phelaomenon was made by SSrensen (1) who was able to show in the case of a number of enzymes ...
